Introduction of plaque assay which is most widely used for titration of influenza virus particles

 

 

 

실험 방법

1. 실험 과정

1) 하루 전, 6 well plate에 MDCK cell을 10⁶ culture를 만들어 둔다.

 

2) micro tube 10^-1 부터 10-6까지 적어준 후 micro tube MEM 900㎕을 각각 넣어준다.

3) virus 100ul을 섞어10^-1 sample을 만들고 10-1에서 100㎕을 따서 10^-2에 넣어주어 10^-2 sample을 만드는 과정을 모두 해준다.

 

4) 6 well plate에 10^-1 부터 10-6까지 적어준다.

 

5) 6 well plate를 PBS로 washing 해 준다.

 

6) 6 well plate에 virus를 각각 넣는다.

 

7) 45분 동안 실온에서 기다린다.

 

8) 6 well plate를 PBS로 다시 washing 해 준다.

 

9) Overlay(2×DMEM 10㎖ + 2% agar + trypsin 80㎕)를 제조한다.

 

10) 제조한 Overlay를 3㎖씩 6 well plate에 넣어준다.

 

11) 37℃ 배양 3일 후 crystal violet staining을 한다.
